Adrenergic drugs are medications that act on adrenergic (adreno-) receptors . Adrenoreceptors are normally stimulated by adrenaline and noradrenaline.
Adrenoreceptors and their effects
Receptor Main organ Main Effect Mechanism α1 Blood vessels Smooth muscle contraction, vasoconstriction, and mydriasis Gq → ↑IP3/DAG → ↑Ca²⁺ α2 Central nervous system Decreases sympathetic outflow Gi → ↓cAMP β1 Heart Increases heart rate & contractility Gs → ↑cAMP β2 Lungs Bronchodilation and smooth muscle relaxation Gs → ↑cAMP β3 Bladder Lipolysis and bladder relaxation Gs → ↑cAMP
Sympathetic agonists (sympathomimetics)
Agonists Examples α1 Agonists Phenylephrine, methoxamine, oxymetazoline, tetrahydrolozine, and xylometazoline α2 Agonists Methyldopa , clonidine , guanabenz, guanfacine, tizanidine, medetomidine, dexmedetomidine, and lofexidineβ1 Agonists Dobutamine, xamoterol (partial agonist), denopamine β2 Agonists Salbutamol, formoterol, salmeterol, terbutaline, and fenoterol β3 Agonists Mirabegron Indirect acting agonists Amphetamine, cocaine , imipramine, tyramine, metaraminol, ephedrine, prophylhexedrine, and 5-methoxy-a-methyltryptamine Mixed agonists Adrenaline (α1, α2, β1, β2, β3), noradrenaline (α1, α2, β1), dopamine (Dopamine + α/β), and isoproterenol (β1 + β2)
Sympathetic antagonists (sympatholytics)
Antagonist Examples α1 Antagonists Prazosin, doxazosin, terazosin, and timazosin α2 Antagonists Yohimbine, idazoxan, and atipamezole α1 and α2 Antagonists Phentolamine, tolazoline, phenoxybenzamine, and dibenamine β1 Anatgonists Atenolol, metoprolol, esmolol, acebutolol, betaxolol, practolol, nebivolol, and bisoprolol β2 Antagonists Butoxamine β1 and β2 Antagonists Propranolol, pindolol (partial agonist-antagonist), oxprenolol, sotalol, timolol, and nadolol Mixed antagonists (α1, β1 and β2) Carvedilol and labetalol
